Virginia Madsen est née le 11 septembre 1963 à Chicago, dans l'Illinois.
Avec 75% de sang italien et 25% de sang irlandais, Gina, comme la surnomment ses proches, est un mélange réussi entre la méditerranée et le nord.
Elle a grandi dans un milieu artistique. Sa mère, Elaine Madsen, était écrivain et poétesse, et son frère, Michael Hudson, est également acteur (Kill Bill volume 2, Blueberry , Die another day entre autres).
Ce fut l’une des stars les plus en vue au milieu des années 80 ; en 1986, elle fut d’ailleurs élue parmi les plus belles actrices.
En 1984, elle interprète la Princesse Irulan dans Dune de David Lynch.
