C'est à l'adolescence que Kathleen Turner se lance dans la comédie, en prenant des cours dramatiques à Londres. De retour aux Etats-Unis, elle s'illustre aussi bien à Broadway que sur petit écran. En 1981, elle fait ses débuts au cinéma en décrôchant le rôle féminin principal du thriller La Fièvre au corps, réalisé par Lawrence Kasdan.
L'année 1985 marque un tournant pour la comédienne, qui se révèle au grand public avec le film d'aventure A la poursuite du diamant vert, où elle donne la réplique à Michael Douglas. L'année suivante, elle s'illustre dans sa suite, Le Diamant du Nil, ainsi que dans L' Honneur des Prizzi, aux côtés de Jack Nicholson.
